What is Dermorphin (High-Dose)??
Dermorphin (CAS 77614-16-5) is a naturally occurring heptapeptide opioid first isolated from the skin of South American frogs (Phyllomedusa sauvagei). Sequence: H-Tyr-D-Ala-Phe-Gly-Tyr-Pro-Ser-NH₂. Molecular formula C₄₀H₅₀N₈O₁₀, MW 802.88 g/mol. This 10mg/vial high-dose format supports dose-escalation studies, multi-arm MOR pharmacology experiments, and long-duration in vivo analgesic paradigms requiring larger quantities. Dermorphin exhibits approximately 1,000-fold greater µ-opioid receptor (MOR) potency than morphine in preclinical binding assays, with high selectivity for MOR over δ and κ subtypes. The D-Ala² residue confers DPP-IV and protease resistance, extending half-life relative to linear endogenous opioids.
Research Applications
High-Dose MOR Pharmacology
Dose-escalation µ-opioid receptor occupancy, Gi/o signalling kinetics, receptor saturation profiling
Analgesic Dose-Response
Multi-arm hot-plate/tail-flick/formalin models across concentration range — morphine comparison
Tolerance & Sensitisation
Repeated high-dose MOR stimulation, receptor internalisation, GIRK channel desensitisation
β-Arrestin & Biased Agonism
β-arrestin-2 recruitment, ERK1/2 phosphorylation bias vs. G-protein signalling at elevated concentrations
D-Amino Acid Stability
DPP-IV resistance profiling — D-Ala² contribution to plasma and tissue half-life
