What is ACE-031??
ACE-031 (also known as Ramatercept, CAS 1621169-52-5) is a recombinant fusion protein composed of the extracellular ligand-binding domain of activin receptor type IIB (ACVR2B) linked to the Fc region of human IgG1. It functions as a soluble decoy receptor, sequestering myostatin (GDF-8), activin A, activin B, GDF-11, and BMP-9 in circulation — preventing these negative regulators from binding membrane-bound ACVR2B on muscle and bone cells. Developed by Acceleron Pharma, ACE-031 has a half-life of approximately 10–15 days in human pharmacokinetic studies.
Research Applications
Myostatin / ACVR2B Signalling
Myostatin-SMAD2/3 pathway suppression, receptor downregulation, ligand-trap kinetics
Skeletal Muscle Biology
Fiber cross-sectional area hypertrophy, anabolic protein synthesis, DMD & ALS wasting models
Bone Metabolism
Biomarker shifts in bone formation vs. resorption, lumbar spine mineral density studies
Fat Metabolism
Adipose lipolysis changes associated with ActRIIB ligand inhibition
Pharmacokinetics
AUC(0-∞), Cmax linear dose-response, T½ 10–15 day profiling
