What is 5-Amino-1MQ?
5-Amino-1MQ (5-Amino-1-methylquinolinium — NNMT inhibitor) is a Small molecule (C₁₀H₁₀N₂, MW 174.2). Competitive NNMT (nicotinamide N-methyltransferase) inhibitor preventing nicotinamide from being diverted away from the NAD+ salvage pathway. Preserves SAM methyl donor pool and improves SAM/SAH ratio for epigenetic methylation. Indirect NAD+ increase → SIRT1 activation → PGC-1α → mitochondrial biogenesis.
Research Applications
NNMT Inhibition
Competitive enzyme inhibition — NAD+ precursor preservation
Indirect NAD+ Upregulation
Salvage pathway restoration vs direct NR/NMN supplementation
Adipogenesis Suppression
PRDM16 methylation preservation → white vs brown adipocyte fate
SIRT1/PGC-1α Axis
NAD+-dependent sirtuin activation and mitochondrial biogenesis
SAM/SAH Methylation
Epigenetic methylation biology and methyl donor pool research
