What is Melatonin??
Melatonin (N-Acetyl-5-methoxytryptamine, CAS 73-31-4) is an endogenous indole neurohormone synthesised from tryptophan → serotonin → N-acetylserotonin → melatonin in the pineal gland, with peak release in darkness. Molecular formula C₁₃H₁₆N₂O₂, MW 232.28 g/mol. Melatonin acts on MT1 and MT2 G-protein-coupled receptors (Gi-linked, cAMP suppression) to entrain circadian rhythms. It is also a direct free radical scavenger (hydroxyl, peroxynitrite, superoxide), upregulates glutathione peroxidase and superoxide dismutase, and participates in immune modulation and mitochondrial protection at the cellular level.
Research Applications
Circadian Rhythm Research
MT1/MT2 receptor activation, SCN entrainment, phase shifting, jet-lag and shift-work models
Antioxidant Biology
Direct ROS/RNS scavenging, GPx/SOD upregulation, lipid peroxidation inhibition
Sleep Architecture
NREM/REM sleep modulation, sleep onset latency, Zeitgeber synchronisation studies
Oncostatic Research
MT1-mediated oestrogen receptor cross-talk, tumour proliferation inhibition in preclinical models
Mitochondrial Protection
Electron transport chain protection, mitochondrial membrane integrity, cytochrome c release
