What is PE-22-28?
PE-22-28 (Spadin analog — TREK-1 potassium channel blocker) is a 7-amino acid synthetic spadin-derived peptide. Synthetic spadin-derived peptide blocking the TREK-1 (TRAAK-related potassium channel-1) two-pore-domain K⁺ channel, which is overexpressed in depression models and reduces neuronal excitability. TREK-1 blockade increases firing rate and serotonin release — antidepressant mechanism without SSRI profile or sedation.
